Principles: COULTER DxH Cell Lyse is a cyanide-free reagent. Because the reagent composition does not contain cyanide ions, the oxyhemoglobin methodology is used. In this method, surfactants are used to lyse the erythrocytes and release the hemoglobin. The released hemoglobin (oxyhemoglobin) reacts with an antioxidant and a stabilizing reagent, converting the hemoglobin into a stable hemochromagen, which is quantitatively measured at an absorbance wavelength.
