Coulter® DxH Cell Lyse, 1 x 5 Liter. Intended Use: Erythrocyte lytic agent for quantitative determination of hemoglobin, enumeration of NRBC, and counting and sizing leukocytes on UniCel® DxH 800 Coulter® Cellular Analysis System. Use only with COULTER® DxH Diluent. Summary: COULTER DxH Cell Lyse, a cyanide-free reagent, is used as a lytic agent to prepare a diluted whole-blood sample for the quantitative determination of hemoglobin and the enumeration of leukocytes and NRBC. Principles: COULTER DxH Cell Lyse is a cyanide-free reagent. Because the reagent composition does not contain cyanide ions, the oxyhemoglobin methodology is used. In this method, surfactants are used to lyse the erythrocytes and release the hemoglobin. The released hemoglobin (oxyhemoglobin) reacts with an antioxidant and a stabilizing reagent, converting the hemoglobin into a stable hemochromagen, which is quantitatively measured at an absorbance wavelength. The lytic reagent solubilizes the red blood cell membrane while simultaneously preserving the white blood cell and NRBC cellular morphology to be counted and sized. UOM: 1/EA.
